Born on Arrakis, to Liet-Kynes and Faroula, Chani is a Fremen. Like all Fremen, she bares the blue in blue eyes, which shows her addiction to spice. Before Paul's arrival on Arrakis, he had visions of Chani on his homeworld of Caladan. When she met Paul, her uncle, Stilgar, instructed her to watch over him. Paul and his mother, the Lady Jessica, became accepted members of Sietch Tabr, where Chani and Paul grew close to one another. They became lovers and had a son, Leto I. Even though Chani was a warrior before Paul and Jessica's arrive, when she learns the weirding way from Jessica and Paul, she became deadly. She used her newly acquired skills to kill the Fremen who desired to call Paul out, who now is a religious leader among the Fremen.

Paul is viewed as a prophet in the eyes of the Fremen. They feel that Paul Muad'Dib is the one to make a paradise out of their desert planet. When Paul drinks the unconverted Water of Life, he is taken into a deep trance. It is only Chani who is able to bring him back to reality. When Paul Muad'Dib is awaked, he has been given the memories of the men and women of his ancestry, making him the Kwisatz Haderach.

When Paul attacks the Emperior and Harkonnen troops, Leto I is killed in the battle by the Emperor's troops. A grief-stricken Chani then had to watch her lover, Paul, marry Princess Irulan to become the Emperor of their universe. However, Paul promises Chani that only she will have his heart and Irulan will only have his name.

At the request of the Bene Gesserit, Irulan fed Chani contraception, which caused Chani to change to a Fremen diet, in hopes to become pregnant. The diet caused her pregnancy with her twins to be accelerated. When she gave birth to Leto II and Ghanima, she lost her life.
